Loss of function and gain of function mutations in GPR54 gene are associated with hypogonadotropic hypogonadism and central precocious puberty respectively. This study was designed to evaluate mutations of GPR54 gene in familial precocious puberty in Iran for the first time. Methods: This clinical-experimental study was held on 225 pediatric patients with precocious puberty who referred to the pediatric department of Imam Reza hospital (2009-2010). Among them, Twenty five pediatric patients who had central precocious puberty with positive familial history genomic were selected. Blood samples were taken for assessment. DNA was extracted and by the means of polymerase chain reaction (PCR), it was amplified and sequenced directly. Data were analyzed by using of Sequencher software. Results: In this study no mutations were found in GPR54 gene while three types of polymorphism: rs10407968 (c.24A>G) in 52% of patients, rs3050132 (c.1091T>A) in 64% and a novel polymorphism c.492C>G in one patient (4%) was distinguished. Conclusion: Polymorphism was found in maximum of the patients with central precocious puberty in this study.}, keywords = {Familial precocious puberty,GPR54gen,Mutation,Polymorphism}, title_fa = {ارزیابی جهش های ژن GPR54 در دختران ایرانی با بلوغ زودرس فامیلیال}, abstract_fa = {مقدمه: مسیر انتقال پیام GPR54/Kisspeptin به عنوان دروازه و تنظیم کننده فعالیت نورون های آزاد کننده گونادوتروپین و عامل کلیدی شروع بلوغ می باشد. جهش های از دست دهنده عملکرد در ژن گیرنده متصل شونده به پروتیین GPR54 )G با تأخیر بلوغ مرکزی و جهش های کسب کننده عملکرد این ژن با بلوغ زودرس مرکزی همراهی دارد. مطالعه حاضر برای اولین بار در ایران و با هدف شناسایی جهش ها در بلوغ زودرس مرکزی به خصوص با موارد فامیلیال انجام شد. روشکار: در این مطالعه آینده نگر که در سال 1388-83 انجام شد، 225 کودک با بلوغ زودرس مراجعه کننده به درمانگاه غدد و متابولیسم کودکان بیمارستان امام رضا )ع( مورد ارزیابی بالینی و آزمایشگاهی قرار گرفتند و از بین آنها، 25 کودک با بلوغ زودرس مرکزی که سابقه فامیلی بلوغ زودرس در بستگان درجه یک یا دو داشتند، انتخاب شدند. نمونه خون بیماران جهت ارزیابی گرفته شد و DNA استخراج شد و به کمک تکنیک واکنش زنجیره ای پولیمراز )PCR( تکثیر و تعیین توالی گردید و نتایج به دست آمده با استفاده از نرم افزار سکوانچر مورد تجزیه و تحلیل قرار گرفت. یافتهها: در این مطالعه، موتاسیونی در ژن GPR54 مشاهده نشد ولی سه نوع پلی مورفیسم مشخص شد: (rs10407968(24A>G در 52% بیماران، (rs3050132(1091T>A در 44% بیماران و پلی مورفیسم جدید 492C>G در یک بیمار مشاهده شد )4%(. نتیجهگیری: پلی مورفیسم ژن GPR54 در اکثر بیماران با بلوغ زودرس مرکزی وجود دارد. }, keywords_fa = {بلوغ زودرس فامیلی,پلی مورفیسم,جهش ژنتیکی,ژن GPR54}, url = {https://ijogi.mums.ac.ir/article_5630.html}, eprint = {https://ijogi.mums.ac.ir/article_5630_a318b3cb298eb3ac1bc312da9c47697c.pdf} }
